HCT116 cells and HT-p21-9 cells were plated at 80000 per well in 96 well plates with indicated agents: 5 nM and 100 nM rapamycin (R5 and R 100), 10 μM LY294002 (LY), UO126, and CGK733, 2 mM NAC. (A) After 4 days, the plates were photographed (upper panel) and lactate concentrations were measured in media (lower panel). (B) An equal volume of attached cells (an aliquot of 2%) was plated in 6 well plates. After 7 days, colonies were stained with Crystal Violet (upper panel) or total numbers of cells per well were determined by counting (low panel).
