Figure 3. Mitochondrial respiration is significantly enhanced in SOX2-high MCF7 cells. MCF7 cells stably-transduced with the SOX2-GFP reporter construct were subjected to FACS sorting to isolate the 5% highest GFP (GFP-high) and the 5% lowest GFP (GFP-low) subpopulations. The Seahorse XF96 analyser was employed to measure the mitochondrial function. In the top panel, a representative OCR tracing is shown. In the lower panel, bar graphs show the basal respiration rate, spare respiratory capacity, proton leak and ATP production, obtained from the OCR quantification. Experiments were performed at least 3 times independently. Results are shown as the mean ± SD: t-test, two tailed-test, *p < 0.05.
