A mouse HCC cell line Hepa1-6 that expresses luciferase reporter was subcutaneously implantated into C57/BL6 mice to generate detectable tumor. From the second week after transplantation, intratumoral injection with pAAV-pCD4-DNMT1, or pAAV-pCD4-scrambled, or pAAV-pCD4-shDNMT1 was done every week till 8 weeks, when the mice were sacrificed.
