(A) The upper panel represents the schematic diagram of constructed plasmid of pGL3/OGT1500, a luciferase reporter plasmid driven by human OGT promoter (−1463 ~ +37). There are six CRE-like elements within human OGT promoter region -1463 ~ +37 bp. pGL3/ OGT1500 or pGL3-basic vector was co-transfected together with pRL-Tk into HEK-293A cells. After 48 h transfection, the luciferase activity was measured and normalized with Renilla luciferase (pRL-TK). The relative activity of luciferase was presented as mean ± S.D., **, p < 0.01. (B) pGL3/ OGT1500 or pGL3-basic vector was co-transfected together with pRL-Tk into HEK-293A cells overexpressing Myc-SIRT1 or Myc-H363Y. After 48 h transfection, the luciferase activity was measured and normalized with Renilla luciferase (pRL-TK). The relative activity of luciferase was presented as mean ± S.D., **, p < 0.01.
