Deglycosylated EpCAM regulates proliferation by enhancing autophagy of breast cancer cells via PI3K/Akt/mTOR pathway

(A, B) MCF-7 cells were incubated with 100 nM Wortmannin or 200 nM Rapamycin for 12 hr after transfected with M-EpCAM plasmid. Expression of PCNA was detected with the method of Western blot. (C, D) MDA-MB-231 cells were incubated with 100 nM Wortmannin or 200 nM Rapamycin for 12 hr after transfected with M-EpCAM plasmid. Expression of PCNA was detected with the method of Western blot. (E, F) MCF-7 and MDA-MB-231 cells were incubated with 100 nM Wortmannin or 200 nM Rapamycin for 12 hr after transfected with M-EpCAM plasmid. The cells were cultured for another 4 days. The CCK8 assay used to evaluate the proliferation of the cells after transfection with the M-EpCAM plasmid or autophagic regulator.
