SRT2183 impairs ovarian cancer by facilitating autophagy

Figure 1. SRT2183 suppresses cell proliferation and triggers apoptosis in ovarian cancer cells. (A) A2780, OVCAR-3, Caov-3, SW626, and SK-OV-3 cells were treated with vehicle or SRT2183 (0.2, 1, 5, and 25 μM) for 24, 48, and 72 h, the cell proliferation was detected by CCK8 assay. (B) A2780, OVCAR-3, Caov-3, SW626, and SK-OV-3 cells were treated with vehicle or 1 μM SRT2183 for 14 days, the cell growth was determined by colony formation analysis. (C) OVCAR-3 and A2780 cells were treated with vehicle or 1 μM SRT2183 for 24 and 48 h, cell apoptosis was detected with the help of Annexin V, propidium iodide staining and flow cytometry. (D) OVCAR-3 and A2780 cells were treated by the same procedures described in (C), the expression of cleaved-Caspase-3 (cleaved-Cas3), and cleaved-PARP was detected by immunoblot analysis. (E) OVCAR-3 and A2780 cells were treated by the same procedures described in (C), the expression of BAX, Mcl-1, Bcl-2 and Bak was detected by western blot.
